Xinhua News Agency, Washington, January 19 - The resistance of pathogens to antibiotics has become an increasingly serious global public health problem. US researchers have recently reported that they have used "genetic scissors" to develop a new system that can determine which genes a particular antibiotic can target for pathogens and is expected to improve existing antibiotics or develop new antibiotics.

Recognized as "genetic scissors," CRISPR gene editing technology precisely locates and cleaves genetic loci on DNA (deoxyribonucleic acid), shutting down a gene or introducing new gene fragments.

Researchers at the University of Wisconsin-Madison recently reported in the British journal Nature Microbiology that CRISPRi is a weakened version of "genetic scissors" that cannot cut DNA strands but attach to a certain position in DNA to prevent it. The protein molecules required for gene transcription are close to each other in order to reduce the gene expression and reduce the number of proteins encoded by the gene. The new system they developed was named "Mobile CRISPRi" and can be used to study different strains.

The researchers found that using this gene editing technique to reduce the amount of protein that is being used as a "target" by an antibiotic, the bacteria become more sensitive to the antibiotic, demonstrating the association between specific antibiotics and certain genes. In this way, researchers can screen thousands of genes that may become potential targets for antibiotics at a time, helping scientists understand the workings of antibiotics and improve drug outcomes.

The researchers used "Mobile CRISPRi" to study Vibrio cholerae isolated from cheese skin to find out how the bacteria colonize the cheese and affect flavor. Researchers say "Mobile CRISPRi" can be used to study any number of pathogens or beneficial bacteria that scientists have not previously known.
